  • Dynabeadsはアセトンに対して耐性がありますか?
  • Dynabeadsの粒子自体は、アセトンを含むいくつかの有機溶媒に耐性があります。
    しかしながらDynabeadsの表面に結合している抗体やストレプトアビジンなどタンパク質への影響はこの限りではございません。

    表面にストレプトアビジンがコートされている場合、アセトンのようなケトン類は避ける必要があります。
    Protein A/Gの場合についてはアセトンに関する情報はありません。

    The beads themselves are compatible with a number of solvents such as: ethanol (70% is actually used to sterilize the beads/washings), methanol, acetone, isopropanol, diglym, DMSO (dimethyl sulfoxide) and DMF (dimethylformamide).
    THF (tetrahydrofuran) is similar to diglym and should work fine with Dynabeads.
    All these will leave the iron oxide precipitates untouched.
    Chlorinated solvents may cause some swelling of the beads.
    Furthermore working with acetonitrile is without problem for the beads, but dichloromethane (CH2Cl2) may release organic polymers from the bead.
    Two important factors for all solvents are concentration and incubation time.
    The surfaces of the different beads gives some restrictions.
    Streptavidin surface: ketones like acetone should be avoided.
    50% DMF is not compatible with streptavidin (it may dissociate into subunits up to 20% might be OK).
    (Please note: the organic solvents will not harm the beads themselves. it is the streptavidin that is the problem)
    Regarding Dynabeads Protein A/G we do not have any information whether these beads are compatible with acetone.
